Multiplexed chemoproteomics for biological insights and functional probe discovery

Abstract -  

Covalent small molecules targeting cysteine residues play critical roles in cellular regulation and drug discovery. The success of covalent drugs against previously “undruggable” proteins has fueled interest in this modality. However, proteome-wide assessment of cysteine ligandability remains challenging due to the vast chemical space and the formidable complexity of the human proteome. We present a multiplexed chemoproteomic platform enabling high-throughput, parallel interrogation of global cysteine ligandability, routinely quantifying ~30,000 reactive cysteines. Applying this platform to screening electrophile libraries, we identified functional molecules that perturb the ubiquitin-proteasome system, revealing new opportunities for therapeutic intervention.
